The Biocompatibility of the Temperature Sensitive pHsp-HSV-TK/As_2O_3 Magnetic Complex and Its Anti-tumor Effect on HepG2 cells

We prepared PEI-As_2O_3/Mn_(0.5)Zn_(0.5)Fe_2O_4 magnetic complex as drug and pHsp-HSV-TK gene vector at the same time it could induce the gene expressed by heat. Then the MTT assay, hemolysis test, the mouse of LD_(50) test and micronucleus (MN) test were presented to evaluate the toxicity of it in vitro and in vivo. The therapeutic effects on cultured HepG2 cells treated by thermochemotherapy and pHsp70-HSV-TK of nanosized PEI-As_2O_3/Mn_(0.5)Zn_(0.5)Fe_2O_4 complex were evaluated by MTT test and flow cytometry. The experiments showed that the toxicity of the material on mouse fibroblast (L-929) cell lines was between 0～1 degree; It has no hemolysis activity; Its LD_(50) arrived at 3.449g/kg; Micronucleus test showed it has no genotoxic effects either. The therapeutic results in vitro showed that thermochemotherapy combined with gene therapy of PEI-As_2O_3/Mn_(0.5)Zn_(0.5)Fe_2O_4 complex could significantly inhibit cellular growth and obviously induce cellular apoptosis. Furthermore, the therapeutic results outweigh that of single As_2O_3 group and Mn_(0.5)Zn_(0.5)Fe_2O_4 group.
