Current study focuses on production and characterization of BC from different cellulose producing bacteria viz., Acetobacter aceti MTCC 2623, Acetobacter liquefaciens MTCC 3135 and Gluconacetobacter xylinus MTCC 7795. Role of media components and various process parameters on growth rate and BC production were studied and optimized. FTIR analysis showed that the characteristic vibrational modes of BC produced were almost same in the typical fingerprint regions as in commercial cellulose. Crystallinity ratio (1,430 cm~(-1)/892 cm~(-1)) varies with media composition and time, proposing their role on spatial arrangement of BC, showing BC to be more crystalline than commercial cellulose. DSC analysis showed that BC produced by A. aceti has glass transition temperature (193 °C) in comparison to A. liquefaciens (73 °C). Bacterial cellulose beads, produced by A. aceti, under defined bioprocess conditions, were used as carrier material for growth and proliferation of Vero cell lines. Growth kinetics of cell lines were analyzed using MTT assay.
