Water soluble polymers used for the delivery of platinum drugs to tumor tissue, vitamin mediated polymer-drug targeting, and hydrogel nanoparticle aggregates as depot controlled-release devices are discussed. Platinum (II) complexes have been bound to poly(hydroxypropyl-methacrylamide) (HPMA) based materials, and superior tumor growth inliibition in mice was observed when compared to the small molecule analogs. Attachment of vitamin Bi2 or folic acid targeting agents to poly(HPMA-co-HPMA-GFLG-methotrexate) conjugates gave superior tumor growth inhibition relative to the untargeted polymer conjugates. Narrow polydispersed hydrogel nanoparticles were synthesized with actives (small molecules or macromolecules) trapped within or between the individual particles of the aggregate network. Data are presented which show that erosion and degradation rates of the aggregate networks can be tailored to control the release of the entrapped molecules.
