We examined in more detail the enhancement of spontaneous excitatory transmission by PAR-1 agonists by applying the whole-cell patch-clamp technique to substantia gelatinosa neurons in adult rat spinal cord slices. PAR-1 agonist peptide TFLLR-NH2 did not affect the peak amplitudes of monosynaptic Aδ-fiber and C-fiber evoked excitatory postsynaptic currents. This result suggests that PAR-1 activation in glutamatergic interneuron terminals results in enhancing the spontaneous release of L-glutamate from there.
