A high-throughput HPLC-ESI-MS/MS methodology has been developed for the simultaneous quantification of two BD-mercapturic acids, MHBMA and DHBMA, in smoker urine. The method has an LOQ value of 0.5 and 10 ng/ml for MHBMA and DHBMA, respectively and requires only 200 ul of urine. The new methodology was successfully applied to a multi-ethnic study examining BD metabolism in European American, Hawaiian, and Japanese smokers (200 per group). Significant differences in the mean urinary normalized MHBMA and metabolic ratios were observed among the three ethnic groups, with the highest MHBMA levels found in European American smokers, followed by Native Hawaiians and Japanese American smokers These results provide preliminary evidence that ethnic groups differ in respect to metabolic activation and detoxification of 1,3-butadiene These differences in metabolism may contribute to the observed variations in lung cancer risk in these populations.
