Purpose: To determine the changes in conformational dynamics of Human a1- Acid Glycoprotein (AGP) upon sequential trimming of the native glycan structures via H/D amide exchange (HDX) coupled with mass spectrometry. Results: HDX-MS has detected changes in the conformational dynamics of AGP as a function of specific glycan structure. Peptide-level HDX reveals that the conformational dynamics in six regions of AGP are affected by different degrees of glycosylation, confirming the potential of HDX to reveal PTM-induced protein conformational changes.
