Time between DBS sample collection and extraction process using tube-based DBS format was significantly shortened by about 2 hours using the microwave heating protocol and could be applied to the analysis of six protease inhibitors. Parallel-micro UHPLC-MS/MS system using low pressure gradient approach requires only two pumps and can be easily converted to column-switching setup. Parallel-micro UHPLC-MS/MS system can accelerate sample throughput and reduce the analysis cycle time by half using two columns without compromising chromatographic separation. MS source performance had no significant loss in sensitivity when operating in the flow rate range of 50-300 μL/min.
