Quantitation of Bacterial Ribosomal Proteins by LC-MS~E

Using an additional dimension of gas-phase separation through ion mobility and multiple endoproteinase digestion (trypsin, chymotrypsin, Asp-N, and Glu-C), quantitation of proteins is dramatically improved. These improvements in quantitation are largely due to increased number of peptides identified and enhanced sequence coverage of proteins. While several low molecular weight proteins tend to generate too few peptides to be quantifiable by this approach even using IMS and multiple enzymes, overall more than 90% of the proteins can be accurately and reproducibly quantified by this improved LC-MS~E.
