Using a quantitative label-free differential proteomics technique, we have compared three human plasma protein targeted antibody depletion methods for their efficiencies to remove targeted high-abundance proteins from monkey plasma. Preliminary results have shown good specificity and reproducibility under optimized loading conditions. Enrichment of non-targeted proteins has been achieved using these depletion methods, resulting in detection of monkey plasma proteins at lower concentration than the undepleted whole plasma samples. For example, Insulin-like growth factor-binding protein 6, Adiponectin, Cystatin-C, Cysteine-rich secretory protein 3, and mannan-binding lectin serine protease 1 were enriched significantly and detected via a one-dimensional LCMS/MS analysis.
