DIGE-directed MudPIT analysis increased the breadth of discovery afforded by combining these two complementary platforms. As expected, MudPIT revealed several additional proteins that were differentially expressed in response to PTEN loss independently of treatment with BKM120, particularly large proteins that were not resolved on the 2-D gels. Also expected, several intact proteins identified by DIGE-MS as changing by charged post-translational modification were not observed as altered in expression based on the peptides analyzed by MudPIT. The current validation studies targeted PP1 alpha, and found several modifications that may be PTEN-dependent. Future studies will investigate PP1 beta and related proteins.
