Accurate; de novo sequencing accuracy reaches 100percent where longest sequences (>100 AA) are assembled. Average sequencing accuracy is 95percent and improves to 97percent when first and last two residues of every sequence are removed. Yields the most accurate MS/MS de novo reconstructions compared to any other automated approach. Robust; high amino acid prediction accuracy is improved from SPS and it is not affected by large numbers of modified peptides. The presence of multiple proteins in the sample does not significantly degrade accuracy. Extensive; generates the longest de novo sequences to date directly from MS/MS analysis without requiring manual annotation or any sequence homology steps. Significant; more can be gained from the protein sample before comparing against a database.
