A rapid and sensitive LC-ESI-MS/MS methods for the determination of goserelin in rat and rabbit plasma were developed and validated. Plasma samples were acidified using hydrochloric acid and were then loaded onto SPE cartridges, which were used to isolate and concentrate the goserelin. As a result of this improved sample preparation, the adsorptive loss of goserelin and interferences by endogenous components of plasma samples were reduced. Under the optimized HPLC and MS/MS conditions, goserelin and an internal standard were well separated, and sharp peaks were obtained with high sensitivity within 5 min. The established methods were suitable for the determination of goserelin in rat and rabbit plasma and were satisfactorily applied to a pharmacokinetic study.
