Quantification of Poly(ADP-ribose)-Binding Proteins Using Multiple Reaction Monitoring (MRM)

More than 300 pADPr-binding proteins were identified including 14 proteins from the NHEJ complex. A MRM method was developed and optimized for the quantitative analysis of the NHEJ proteins. The effect of MNNG on the NHEJ proteins bound to pADPr was measured by MRM. 10 NHEJ proteins were detected and their concentration was higher after 5 min of MNNG treatment compared to the untreated control.
