Matrix Optimization for MALDI-TOF MS Lipid Fingerprinting of Single Bovine Embryos

MALDI-MS fingerprinting has been recently shown to provide reproducible lipid profiles of single and intact cocytes and embryos using DHB as matrix in the positive ion mode. The profiles were also influenced by different bovine embryo in vitro culture systems [J. Lipid Res. 2010 51 (5), 1218-1227]. The objective of this work is to evaluate MALDI-MS lipid fingerprinting from single bovine embryos in the positive and negative ion modes using three different matrix compounds recently reported for improved lipid analysis. These new matrices may enhance the quality of MALDI-MS single embryo fingerprinting most particularly for monitoring embryo development and storage. Also, performing analysis in both negative and positive ion modes can be advantageous regarding the types of lipids detected.
