Despite the overwhelming clinical significance of metastases, the cellular and molecular mechanisms involved are largely unknown. By analyze function and protein network of differentially expressed proteins among metastatic cancer, we could expand our knowledge about cancer metastasis. The object of our study is define significant differences between primary colon carcinomas and their metastases. To address the object, we obtained normal colon tissue, primary colon carcinoma and their matched metastatic carcinoma from the liver from 2 metastatic colon cancer patients, in which one person was a synchronous case in which primary and metastatic tumors were found at the same time, and the other person was a metachronous case in which metastatic tumors were found after surgery on primary tumors. Proteins extracted from each tissue section was labeled with iTRAQ~R reagents - 8plex, and analyzed with multi-dimensional liquid-phase peptide fractionation followed by tandem mass spectrometry. Thousand proteins were confidentially identified in cancer tissue. Especially, as cancer was developed many proteins were changed in metastasized tissue compared to original primary cancer tissue. Our profiles of the proteomic alterations of the primary colon cancer and its transition to metastasis may provide a better understanding of the molecular biology and insight into tumor progression and metastasis with the aim of identifying biologically and clinically meaningful target proteins.
