An LC-MS/MS method has been validated for the quantitation of itraconazole (1-500 ng/mL) and hydroxyitraconazole (2-1000 ng/mL) in K_(2)EDTA human plasma using liquid-liquid extraction. The method is sensitive, selective, accurate, and reproducible. Both itraconazole and hydroxyitraconazole are stable after five freeze/thaw cycles, six hours storage at room temperature. This method provides a useful tool in quantification of itraconazole and hydroxyitraconazole in pharmacokinetic and bioequivalent studies.
