Ranging from bacteria to mammals, protein secretion has been established as a fundamental cellular process subserving a wide range of biological functions. In the brain, protein secretion plays an important role throughout an organism's lifespan, providing cues that support synapse formation during development and coordinate responses to inflammation. Identification and quantification of these secreted proteins is a first-step in understanding their roles in biological and pathological processes. In addition, these proteins can be used as biomarkers for disease diagnosis or reveal key proteins that participate in disease progression. Towards these goals, we utilized SILAC in primary murine astrocytes to generate isotope-labeled reference proteomes for quantifying dynamic changes in the intracellular and extracelluar proteomes.
