LAESI method successfully used for direct in situ detection of metabolites and lipids from cells. HTLV-1 infected cells show profound changes in the relative quantities of metabolites and lipids compared to healthy cells (C81 vs. CEM); similar changes are found in H9-Tax vs. H9 cells (data not shown). Choline, glutathione, spermine, and phosphocholine (among others) are down regulated in HTLV-1 infected T cells, whereas AMP, arginine and creatine levels are upregulated. Future directions: 1. Application to other viral infections, such as HIV, KSHV, etc. 2. Measurement of absolute quantities of selected metabolites using deuterium labeled standards.
