In conclusion, three LC/MS/MS bioanalytical methods with SLE sample preparation and fast liquid chromatography were developed and validated for the determination of BILR 355 and its isobaric metabolite BILR 516 in human plasma. In these separations, baseline separation of the two analyte/IS pairs is critical for method performance. These fast chromatographic assays demonstrated that the amaller particle size (1.8 and 2.5 (mu)m) columns used with high flow rate produced comparable accuracy and precision, but much narrower peak width and better sensitivity over conventional HPIC without sacrificing chromatographic peak shape yet achieving much shorter run times.
