Developed mass spec based epitope mapping method to locate the region on AM that binds to antibodies, validated by SPR biosensor technology. Developed affinity capture LC-MS/MS method to quantify AM from serum. (1)The nano system achieved a LOQ of 200 amol (6 pg/mL serum). (2)The capillary system achieved a LOQ of 1000 amol (30 pg/mL serum). Developed method to quantify AM bound to CFH. (1)AM concentration is approx3 nM in 0.5 mg/mL commercial CFH samples. (2)Binding stoichiometry approx 1:1000.
