Identification of a Novel Metabolite for The HIV treatment Tenofovir Disoproxil with LC-MS/MS

Tenofovir Disoproxil Fumarate (TDF) is a nucleotide analogue reverse transcriptase inhibitor, the active form is tenofovir diphosphate. Whilst developing a rapid LC-MS/MS method to quantify systemic levels of the HIV drug treatments TDF, Emtricitabine and Efavirenz in human plasma a novel metabolite of TDF, not previously reported in humans was detected. The method was based on reversed-phase (RP) liquid chromatography (LC), on a Waters Acquity UPLC using a 2.1 mm × 100 mm HSS T3 Acquity column with detection via Multiple Reaction Monitoring (MRM) using a Waters XevoTQ-S tandem quadrupole mass spectrometer. Accurate mass experiments were performed on a Waters Q-ToF Synapt G2-S mass spectrometer.
