Native mass spectrometry (MS) has a strong potential for its application towards the systems where other biophysical techniques are futile (1). In yeast, the dedicated guided entry of tailanchored proteins (GET) pathway targets and inserts tail-anchored (TA) proteins into the endoplasmic reticulum (ER) membrane. A complex of two membrane proteins, Get1 and Get2, captures the TA protein from the Get3 cytoplasmic chaperone and mediates its membrane insertion.
