Noncovalent complexes formed between lysozyme and dyes were monitored using MALDI-ToF-MS. The best signals where obtained for the mixture in 1:5 (protein:ligand) molar ratio. The intensities of signals of complexes increase in the order Xylenol Orange, Solochrome Cyanine R, and Lissamine Green. Comparison of the complexes indicated that differences in complex ion formation may be caused by availability of electrolytic groups in their structures. The effect of cationizing agents on complex formation during MALDI was evaluated. It was seen that presence of salts, NaTFA, KTFA, and LiTFA enhanced the intensities of obtained complex signals in mass spectra.
