Laser irradiation is an efficient method to induce protein crosslinks, especially for an engineered non-natural amino acid incorporation, and should be amenable to in-vivo examinations. High resolution MS is a useful tool to investigate and identify the protein crosslinks. It is advantageous to use distinct and complementary informatics approaches for crosslinked peptide identifications, to minimize ambiguity. BpaAF binds specifically to the drug site I in BSA. The use of an unnatural amino acid in Ste2p should afford the ability to examine protein interactions in this GCPR.
