Using experimental design and reference materials (standards), we optimized capillary and fragmentor voltages for LC-ESI-MS analysis. For all alkaloids studied, we observed the higher and more reproducible molecular protonated (MH~+) ion currents at 1000 V-capillary voltage and 150 V or 200 V (monocrotaline) fragmentor voltages. Under these ESI operation parameters, it was possible to detect these alkaloids in a very small plant sample (< 0.1 g), subjected to matrix solid-phase dispersion(MSPD).
