We have investigated an immuno-LC-MS/MS assay for the sensitive (LOD in the ng·mL~(-1) range), rapid (total assay time < 2 hours) and specific detection of two Staphylococcal enterotoxins. Sample preparation (immunoextraction) and chromatography separation were optimized for maximumsensitivity. Addition of labeled protein standards (PSAQ standard) leads to toxin absolute quantification in the samples and also provides a positive control, monitoring the whole analytical process. Application to food and biological matrices is undergoing.
