EXCRETION OF NATIVE YEAST METABOLITES AND THE ROLE OF ABC TRANSPORTERS

Using LC/MS and non-targeted multivariate data analysis to explore yeast secretions, we were able to characterize a subset of secreted compounds to then target for observation in knockout mutant studies. Experimentation with mutants positively implicated ABC transporters in the secretion of a number of these compounds, and using individual transporter knockout mutants, we were able to pinpoint a specific transport protein of interest (PDR5) for future study, as well as 4 of its substrates by mass. Furthermore, we were able to positively identify 6 of the 53 secreted compounds by LC-MS/MS. 4 of these compounds mark new discoveries in secreted yeast metabolites. We find it remarkable that such a well studied system can benefit so readily from the simple application of modern LC-MS technology and statistical analysis. As high resolution LC-MS becomes increasingly accessible to research laboratories, we expect to see even in the most studied systems yield remarkable discovery.
