Linker histones were successfully purified from chicken erythrocytes under native conditions. Mass spectrometry has been to identify a range of post-translational modifications. These results suggest that similar to the core histones, post translational modifications of linker histones may regulate their function in vivo. A large number of the modifications identified on the chicken linker histones in this study differ from those previously observed in linker histones in human cultured cells. It is likely that these differences are related to the different functions of the two cell types. Further studies are required to determine how the PTMs of linker histones correlate in their action with core histone PTMs and nucleosome remodeling.
