A rapid, selective, sensitive, accurate and reproducible method for theophylline was developed and validated. Method robustness was demonstrated through the use of multiple extraction scientists and mass spectrometer platforms. A post-column infusion demonstrated that theophylline peaks did not co-elute with any areas of significant suppression or enhancement. Highly effective and efficient chromatography was developed on a AB|MDS Sciex API 4000 that achieved lower limits of quantitaion for theophylline of 100 ng/mL with an acquisition time of 2.2 minutes. The method was shown to be highly selective in the presence of very similar and isobaric co-administered analytes.
