Tripterygium wilfordii Hook f. (T.wilfordii) is a traditional Chinese medicine. The main active components are triptolide and alkaloids, but a mass of toxic component tripterine often exists in the extract derived from traditional extraction basing on organic solvents. The supercritical fluid extraction (SFE) and liposomes technology were employed to reduce the toxicity and enhance the efficiency of T.wilfordii in this study. The effect of co-solvents, the extraction temperature and pressure, the soak time on the yield and content of the triptolide, alkaloids and tripterine were investigated to obtain the suitable operational conditions. In the liposomes experiments, the processes parameters are studied to reach the high entrapment efficiency of the tripterine and alkaloids. The results show that the yield of triptolide is 3.49 times as much as that of traditional extraction when the co-solvent is 75% ethanol, the extraction temperature and pressure are 43°C and 25MPa respectively and the soak time is 3h. The following liposomes experiments showed that 98.10% and 88.63% of tripterine and alkaloids can be entrapped into liposomes respectively when the film-making temperature is 40°C, the dispersant is phosphate solution containing 0.5mg/mL surfactant Tween-80 and its pH value is 7.6, the mass ratio of extract to lipids and cholesterol to lecithin are 0.2 and 0.25 respectively. The combination of SFE and preparation of liposomes can reduce the toxicity and enhance the efficiency of T.wilfordii.
