The molecular events in Ag-IgE Ab-FceR dependent mast cell exocytosis is fundamental to the role in innate and acquired mast cell immunity. Advanced molecular technology made it possible to realize the formation of Ag-IgE Ab complex, which control the stimulus strength and mast cell effector responses. In contrast to the refined molecular analysis of IgE epitope of antigen, study of variable region structure, which is involved in Ag recognition of IgE, remain unsuccessful. Recently, the entire structure for the gene locus of human Ig heavy chain V-region (VH) have been elucidated. With a new approach using VH family primers, the IgE-VH0repertoire of patients with allergic disorders have been analyzed. It is of interest to inspect IgE-VH0repertoire from young infant because they are exposed to rather limited environmental antigens than adults. Moreover, these findings provide the tools for analysis of the rearrangements in IgE-committed B cells. We therefore employed cDNA-PCR approach to analyze IgE VHtranscripts expressed in PBL from atopic children.
