Hormones and stroma play major roles in prostate cancer progression. The studies presented within these proceedings utilized the technique of tissue recombination to evaluate the roles of steroid hormones and stroma on prostate cancer progression. Testosterone and estradiol stimulated carcinogenesis in tissue recombinants consisting of embryonic prostatic stroma and non-tumorigenic human prostatic epithelial cells (BPH-1). In another model, addition of carcinoma associated fibroblasts to BPH-1 cells stimulated tumor formation and malignant transformation. Currently these are the only models that recapitulate human prostate cancer progression of benign to cancer utilizing factors found naturally in the body. These data suggest a major role for stromal participation in carcinogenesis.
