摘要

A series of poly(ethylene oxide) (PEO) matrices for separation of specific DNA fragments in a PMMA-based chip were quickly developed by using an orthogonal designing approach. The matrices were composed of PEOs in four size ranges (M_w: 8x10~6, 1x10~6, 4x10~5, and 1x10~5) at varying concentration ratios, suitable for analyses of multiplex PCR and digest fragments ranging in size from 20 bp to 40 kbp. In mixed solutions, PEO with intermediate molecular weight (MW) of 4x 10~5 was a dominant factor for separating small DNA pairs (e.g., 82/88 bp), but PEO with high MW of 8x10~6 played an important role in separating other pairs (e.g., 271/281 bp, 506/517 bp, 7 k/10 kbp).
