The present invention relates to methods of producing eukaryotic cell libraries encoding a repertoire of binding molecules ("ligands"), wherein the methods use a site-specific nuclease for targeted cell DNA cleavage to enhance integration. specific site of ligand genes through endogenous cell repair mechanisms. eukaryotic cell populations are produced in which a repertoire of ligand-encoding genes are integrated into the desired locus in the cell DNA (e.g., a genomic locus) allowing expression of the encoded binding molecule thereby creating a population of cells expressing the different ligands.
