For this talk, I am going to focus on pituitary-dependent hyperadrenocorticism (PDH) Mitotane (o, p' DDD Lysodren~R) remains my mainstay of medical therapy for dogs with PDH. Its mechanism of action is to cause a relatively selective necrosis of the adrenocortical zones that secrete Cortisol; secretion of aldosterone is usually preserved. When using mitotane, treatment is divided into two phases: induction (loading) and maintenance Mitotane should always be given with food, since this increases the bioavailability of intact tablets.
