The medium was employed and divided into two factors for further optimization with shaking cultivation on the basis of the previously related research. the relationship between component factors (x_i) of the medium and spore numbers (y) of Bacillus thuringiensis (Bt) recombinant was demonstrated with the central composite rotable design (CCRD). A response surface equation was formed as follows: y=308.5-28.34_(x_1) + 3.17_(x_2) - 30.25_(x_1~2) - 61.25_(x_2~2) + 46_(x_1x_2) and then the best combination of medium, ∑C_1 = 0.75%, ∑C_2 = 0.44% and ∑C_3 = 0.1%, was rapidly acquired.
