Despite current treatments, local GBM recurrence and associated mortality is almost 100%. To reduce local recurrence and overcome the blood-brain barrier, drug delivery locally to the tumor resection site is highly advantageous. Here we explored the novel biodegradable polymer Ace-DEX as a tunable polymer platform for sustained interstitial drug delivery to prevent GBM recurrence. Drug loaded scaffolds exhibited sustained drug release which translated to robust suppression of tumor recurrence and significantly improved overall survival of mice in a clinically relevant murine model of surgical GBM resection and recurrence.
