The aim of this work was to develop a system that candeliver siRNA into cells present in the human epidermis.More specifically, we wanted to block the expression of aspecific Myosin Va exon F containing isoform that isinvolved in melanosome transport in human melanocytes[1]. By knocking down this isoform, we might be able totreat various forms of hyperpigmentation. Therefore, weprepared and investigated the capacity of a new type ofethosome to deliver siRNA into hard-to-transfect humanprimary melanocytes. This works follows from ourprevious published data [2]. Results show that theethosomes are highly deformable, efficiently deliverysiRNA into primary melanocytes and maintain stabilityfor at least 3 weeks. Their penetration capacities throughhuman skin are currently being evaluated.
