The integrity of the blood-brain barrier(BBB) is failed after cerebral ischemia andreperfusion. A further elucidation of thisphenomenon is required to develop a treatmentstrategy for reperfusion injury. In the presentstudy, we determined the time-dependentchange of reperfusion injury and BBB disordersin a rat model of cerebral stroke andreperfusion. For this purpose, fluoresceinisothiocyanate (FITC)-dextran was injected intotransient middle cerebral artery occlusion (t-MCAO) rats. As a result, the leakage of FITCdextranfrom cerebral vessels occurredimmediately after the reperfusion prior to theserious reperfusion damage. The present dataprovides important information for constructinga drug delivery system (DDS) focused on BBBdisorders for the treatment of reperfusion injuryafter cerebral stroke.
