We studied the sensitivity of Sf9 cells to the Japanese B Encephalitis Virus (JEV) in serum and serum-free media, the infection type of JEV in Sf9 cells and the passage stability of JEV in them. At first, the results indicated that JEV could propagate in Sf9 cells in both serum and serum-free media; the maximum virus titers were 6.20 and 4.58 respectively. The infected cells continued to live and maintain high cell concentration. It was concluded that JEV can bring virus-generating continuous infection to Sf9 cells. The second, sub-culturing Sf9 cells for 3 successive generations in serum medium, we got the maximum virus titer 6.27, 6.00 and 6.18 in each generation, and in serum-free medium, the maximum virus titers were 4.55, 4.60 and 4.53, which showed little difference with that of parents viruses. It was proved that JEV has passage stability in Sf9 cells.
