Silica nanoparticles (SNPs) and poly (amido amine) (PAMAM) dendrimers of different generations (G 3.5-G7) with different surface functional groups, sizes, and geometries were systematically evaluated for toxicity in immune-competent CD-I mice after intravenous and oral administration. Previously unreported fatal complication such as DIC, intestinal hemorrhage, and haemobilia were observed. Surface functional group was the most significant factor in predicting the toxicity of these nanoconstructs in vivo.
