Methods are provided for producing hepatocytes and/or cholangiocyte lineage cells from pluripotent stem cells, the methods comprising: (a) converting an extended nodal agonist treatment induced endoderm cell population to an FGF agonist and a BMP4 agonist and/or its characterizing the extended nodal agonist treatment induced endoderm cell population by contacting it with a characterization medium comprising active conjugates and/or fragments to obtain a cell population comprising hepatocytes and/or cholangiocyte progenitors; (b) inducing maturation of hepatocytes and/or cholangiocyte progenitor cells of the cell population, and optionally further lineage characterization and/or expansion to obtain a population comprising hepatocyte lineage cells such as hepatoblasts, hepatocytes and/or cholangiocytes and wherein inducing maturation comprises generating an aggregate of the cell population. Optionally, the method also includes activation of the cAMP pathway within the aggregate and formation of co-aggregates.
