PROBLEM TO BE SOLVED: To provide methods for producing hepatocyte and/or cholangiocyte lineage cells from pluripotent stem cells.;SOLUTION: Methods for producing hepatocyte and/or cholangiocyte lineage cells are provided, the methods comprising the steps of (a) obtain a cell population comprising hepatoblasts by contacting the extended nodal signal transmission agonist treated induced endodermal cell population with specification media comprising a FGF agonist and a BMP4 agonist and/or active conjugates and/or fragments thereof and thereby specifying the extended nodal signal transmission agonist treated induced endodermal cell population ; and (b) inducing maturation of the hepatoblasts, and optionally further lineage specification and/or expansion of the hepatoblasts, whereas the population of the hepatoblasts is dissociated to obtain the aggregates thereof, and the aggregates are at least cultured in a maturation media, optionally in a gelatinous matrix, in the presence of hepatocyte growth factor, dexamethasone and/or Oncostatin M and/or active conjugates and/or fragments thereof.;SELECTED DRAWING: None;COPYRIGHT: (C)2020,JPO&INPIT
