Provided herein are improved methods for robust TCR+ cell depletion and generation of TCR- cell populations that may be beneficial in minimizing the risk of GvHD in patients receiving allogeneic CAR T cell therapy. Provided herein are methods of increasing the efficiency of depleting TCR+ cells from a population of cells to significantly reduce any residual levels of TCR+ cells present in the cell population with reduced or eliminated expression of endogenous TCR. Associated kits and cell populations are also provided.
