Disclosed in the present invention are an immunofluorescence test kit for measuring PD-L1 expression in circulating tumor cells in peripheral blood in a prostate cancer patient, and a measurement method, the test kit comprising a diluent, a decoloring solution, a dyeing solution A, a dyeing solution B, methanol, 2% PFA, 10% goat serum, a primary antibody suspension consisting of mouse anti-CK, rat anti-CD45, and rabbit anti-PD-L1 antibodies, a secondary antibody suspension consisting of fluorescent-conjugated goat anti-mouse, fluorescent-conjugated goat anti-rat, and fluorescent-conjugated goat anti-rabbit antibodies, and a DAPI mounting medium. The measurement method comprises peripheral blood collection, membrane filtration separation, dyeing, determining whether CTCs are present, and measuring PD-L1 expression in CTCs in the peripheral blood by means of immunofluorescence. In the method of the present invention, expression of PD-L1 in a late-stage or relapsed prostate cancer patient can be measured without the use of puncture biopsy to collect a tissue sample, and minimally invasive technology is used, thus being able to implement real-time dynamic measurement.
