The present disclosure generally relates to novel methods for sequencing nucleic acids. Specifically, the present invention relates to a technique based on liquid chromatography-mass spectrometry (LC-MS) for directly sequencing RNA without cDNA. One of skill in the art can simultaneously read RNA sequences with single nucleotide resolution while determining the presence, type and location of a wide range of RNA modifications. The present disclosure directly sequences RNA without the need for prior cDNA synthesis, simultaneously sequences the nucleotides of RNA molecules with single nucleotide resolution, and indicates the presence, type, location and amount of RNA modifications.
