Recombinant recombinant vaccinia Ankara (RMVA) vaccine with improved stability and its preparation method

A vaccine composition comprising recombinant EI vaccinia Ankara (RMVA) virus, which is genetically stable, which is genetically stable at least ten successive generations, including IEI, IE2 and pp65 or their antigenic fragments. A method for improving the stability of such RMVA at successive times, as follows:(1) inserting one or more nucleic acid sequences encoding a CMV antigen or its antigenic fragment into one or more insertion sites not including but not limited to(2) codon optimization of the nucleic acid sequence encoding the CMV antigen by removing continuous cytosine or guanine; AndIntroducing one or more mutations into the amino acid sequence of the CMV antigen;A method by including one or more of the above.
