Pichindevirus reverse genetics system and method of use. Provided herein is a genetically modified pitine devirus comprising three ambisense genomic segments. The first genomic segment includes a coding region encoding a Z protein and a coding region encoding an LRdRp protein. The second genomic segment includes a coding region that encodes a nucleoprotein (NP), and the third genomic segment includes a coding region that encodes a glycoprotein. The second and third genomic segments each include an additional coding region that can encode an antigen or a detectable marker. Also provided herein are reverse genetics systems for making genetically modified pitindeviruses and collections of vectors that can be used to make genetically modified pitindeviruses.
