Disclosed is a catalyst-free electrochemical deuteration method in which deuterium oxide is a deuterium source, the method comprising: adding an electrolyte, an organic substance containing an ethylenic bond or acetylenic bond, deuterium oxide and an organic solvent to a reactor, using carbon felt as an electrode material and applying a 4-8 V direct-current voltage under an inert gas atmosphere for an electrolysis reaction, and purifying the reaction product to obtain a deuterated product. The deuterated product is obtained by means of direct-current electrolysis in an organic solvent using deuterium oxide as a deuterium source and a cheap and readily available carbon electrode material as a cathode and an anode. The reaction process does not require any transition metal catalyst, the yield of the reaction is 50%-90% and the deuteration rate is above 90%. As the use of a transition metal is avoided, the reaction is suitable for the modification of drug molecules in the later stage. In addition, due to the reaction path being different from the reaction process of transition metal catalysis, chemical selectivity different from that of a deuteration reaction catalyzed by a transition metal can be achieved.
