摘要
Enzymes produced by the fermentation of micro-organisms are treated with 0.005-1.5 parts by weight of protamine (based on total N in enzyme material) in 0.01-1.0 N solution of an alkaline earth cation at 0-40 DEG C. and pH 5.0-8.0, whereby the impurities are precipitated. Preferred conditions are pH 7.0-8.0 at 0-20 DEG C., the enzyme and protamine solutions being prepared separately at the same pH and temperature and then mixed; the alkaline earth metal may be in either or both solutions. The solution preferably contains 0.2-2 per cent of protamine and is 0.05-0.1 N as regards the alkaline earth metal. The alkaline earth metal may be calcium, strontium, barium, magnesium or beryllium, supplied as halide, sulphate, phosphate, acetate or citrate. An example is given of the process applied to the purification of a mixture of streptokinase and streptodornase. Other kinases and desoxyribonucleases, and enzymes such as hyaluronidases and collagenases may also be so treated.
